Glyko® Sialidase S™ [GK80021]

Price:
$ 260.00

Description

Neuraminidase, releases α(2-3)-linked sialic acid from oligosaccharides, glycoproteins, complex carbohydrates. Replaces Glyko® Sialidase S™ (GK80020).

Source:
Glyko® Sialidase S™. Recombinant from Streptococcus pneumoniae, expressed in E. coli. 6xHis-tagged.

Specificity:
This enzyme preferentially releases α(2-3)-linked N-acetylneuraminic acid (Neu5Ac, NANA) from complex carbohydrates. Sialidase S is also active against N-glycoylneuraminic acid [1] (Neu5Gc, NGNA), although similarly to other sialidases [2] the activity is lower toward Neu5Gc than Neu5Ac.

Although the α(2-3)-linkage is preferred, Sialidase S (also known as NanB) is also capable of cleaving α(2-6)-linked sialic acid [3], particularly at higher concentrations.  Adding an excess of this enzyme can lead to some cleavage of α(2-6)-linked sialic acid. Using GK80021 as directed in the Certificate of Analysis reduces the likelihood of these effects.  Users may wish to optimize the enzyme concentration needed for specific proteins.

May be used for exoglycosidase sequencing in conjunction with Sialidase A (GK80040), which has a broader specificity, releasing α(2-3,6,8,9)-linked sialic acids.

FAQs

Q. Does GK80021 contain a purification tag, such as 6xHis?

A. Yes. GK80021 contains a 6xHis tag.

Q. The pack size of GK80021 is 3 Units, which you list as equivalent to 1 Unit of previous Sialidase S product GK80020. Why the difference? Has the unit definition changed?

A. The unit definition has not changed.  We are performing the activity assay in a different format (used also for Sialidase A and other sialidases), which produces consistently different values from the previous assay format.

Q. Is sialidase S exclusively specific toward α(2-3)-linked sialic acid? Can it cleave α(2-6)-linked sialic acid residues?

A. Although the α(2-3)-linkage is preferred, Sialidase S (also known as NanB) is also capable of cleaving α(2-6)-linked sialic acid [1], particularly at higher concentrations.  Adding excess of this enzyme can lead to some cleavage of α(2-6)-linked sialic acid. Using GK80021 as directed in the Certificate of Analysis reduces the likelihood of these effects.  Users may wish to optimize the enzyme concentration needed for specific proteins.
