Cleaves between the GlcNAc residues of the chitobiose (GlcNAc-GlcNAc) core of N-linked glycans. Oligomannose & some hybrid type N-glycans are released from glycoproteins, leaving a single GlcNAc attached to asparagine. This specificity is distinct from Endo F2 (GKE-5008) which also cleaves biantennary complex type N-glycans, and PNGase F (GKE-5006) which releases most N-glycans by cleaving between the innermost GlcNAc and asparagine.

Selective release of high mannose N-linked oligosaccharides from glycopeptides/ glycoproteins. The enzyme has found extensive use in the characterization of glycoproteins, dolichol-linked glycans and the biosynthetic pathway for N-glycosylation.

One unit is defined as the amount of enzyme required to catalyze the release of N-linked oligosaccharides from 60 μmoles of denatured ribonuclease B in 1 hour at 37°C, pH 5.5.
